For the traffic aspect, what Google does for Google Maps is it takes around 100 phones on the same road and measures their speed. If they are fast, the road is green. If they are slow, the road is red. Also I'm pretty sure Google uses automated systems for the labels on everything.

Routes are calculated with weighted pathing algorithms like A* (a-star), and fine tuned based on trends, like popular routes and traffic.
